研究方向:
遗传学与表观遗传学,干细胞生物学与发育
研究内容:
印记基因在哺乳动物发育过程中有不可或缺的作用。印记基因调控失常会导致癌症,糖尿。难懿『蜕窬约膊〉刃矶嗳死嗉膊。他的实验室首先发现与印记基因遗传和调控有关的主要因子ZFP57。他们发现ZFP57的缺失会阻碍一个非常重要NOTCH信号通路的激活,进而影响心脏发育。在小鼠突变体里发现的心脏发育缺陷跟由于ZFP57基因突变引起的人类疾病完全一致。这证明了他们用小鼠动物模型来研究印记基因与人类疾病关系的可行性。他的实验室第一次在哺乳动物中发现母源-合子效应。母源和合子的Zfp57基因产物都调节NOTCH信号通路和心脏发育,它们的功能互补但不完全重叠。最近,他们与合作者一起证明ZFP57调控大部分已知印记基因的单等位基因表达和转换机制。目前他实验室主要研究印记基因和NOTCH信号通路在心脏和神经中的功能以及相关人类疾。渲邪ɡ┱判托募〔(DCM)和阿尔茨海默症(Alzheimer’s disease)。 他实验室另一个重要研究方向是干细胞表观遗传学,主要是针对印记基因在多能性干细胞中的遗传与调控。他们的研究成果表明ZFP57在胚胎干细胞(ES)和诱导干细胞(iPS)中对印记基因的正常表达非常重要。诱导干细胞(iPS)和核移植获得的胚胎干细胞(ntES)是将来最有可能被用于阿尔茨海默症等退行性疾病的细胞治疗。许多研究表明印记基因在这些表观遗传重编程获得的多能性干细胞中时常发生表达异常,而印记基因的非正常表达会导致癌症等很多人类疾病。他的实验室在探索一些方法帮助将来获得适合用于细胞治疗的胚胎干细胞和诱导干细胞。
公海彩船: 研究成果展示
我们课题组发现ZFP57是维持基因组印记的主要调控因子,并且调NOTCH信号通路和胚胎心脏发育。ZFP57具有母源-合子效应,这是首次在哺乳动物中被发现。我们证明ZFP57维持印记基因单等位基因表达特性,它的缺失会导致印记基因的等位基因表达发生转换。现正检验ZFP57和它的靶向印记基因通过NOTCH等信号通路调节器官发育、小鼠神经行为及记忆的机理。多年前我们发现干细胞诱导过程会导致基因组印记异常,这不利于干细胞治疗。现正利用基因编辑技术发掘ZFP57、DNA甲基化转移酶和染色体甲基化修饰酶等因子在胚胎干细胞中维持基因组印记的调控机制,为将来获得有稳定基因组印记并适合细胞治疗的干细胞打下基础。
